This invention describes a toll-like receptor knockout animal that is unresponsive to bacterial cell components, namely, peptidoglycan and a lipoprotein/lipopeptide. Further disclosed are methods for elucidating the contribution of individual members of the TLR family to a signaling stimulated with bacterial cell components in vivo, in particular, the role of TLR2 and MyD88 in vivo. Also described are the means by which such a knockout mouse is generated, resulting in a homozygous disruption of the TLR2 gene in its genome, wherein such disruption results in no production of endogenous TLR2 protein, and wherein the resultant mouse exhibits the phenotype of being unresponsive to bacterial cell components such as a lipoprotein and lipopeptide. Applications are included in the screening of potential modifiers for a desired response in pharmaceutical use.
