Described herein are derivatives of tetrahydropyran or cyclohexane, their methods and uses. The invention relates to compounds and compositions useful for the modulation of ligase activity, specifically to inhibit or activate E3 ubiquitin-protein ligase. Further disclosed are methods of use as a preservative of cell, blood, tissue or organs or as an agent to modulate stem cells. Therapeutic applications are provided in the treatment and prevention of neoplastic disorders, acute or chronic renal failure, inflammatory disorders, immune disorders, cardiovascular disease, and various types of cancer.
