This invention describes a novel gene-targeted mouse heterozygous for human Familial Alzheimer’s Disease (FAD) mutation, and the generational offspring thereof, comprising a human mutation of FAD presenilin-1 (PS-1), a human transgene for FAD Swedish APP695, and a humanized Abeta (A?42) mutation, such that the A?42 protein level is elevated relative to the A?42 protein level in a wild-type mouse. Also included are therapeutic applications of such a non-human mammal in identifying compounds for the treatment of Alzheimer’s disease.
