This invention introduces a nonhuman mammal and its progeny with suppressed expression of the gene encoding CD45 on B cells. Specifically, the authors describe a mouse lacking expression of a particular CD45 isoform in certain cells of the immune system, such that the mouse may optionally contain a transgene encoding the CD45RO isoform. Further disclosed is a DNA sequence comprising at least exons 6-8 of a mouse CD45 DNA construct into which a marker gene sequence has been substituted for at least exon 6 and intron 6. Specific DNA sequences are delineated, as are applications in the screening of drugs for their immunomodulatory effects.
