Compound screening assays using a transgenic mouse model of human skin diseases

Longley, B. Jack; Hamden, CT, USA

This invention provides methods of identifying a composition, a compound or a procedure which can either produce a skin response in a subject, or reduce and treat a skin response in a subject. Further provided is a method of identifying a composition, a compound, or a procedure which can reduce radiation damage to the skin of a subject, and a pharmaceutical composition for treating human skin diseases. The model is the skin of a transgenic mouse which has been hyperpigmented and which has stably integrated into its genome a nucleic acid comprised of a human cytokeratin 14 promoter linked to a nucleic acid which encodes the epidermal stem cell factor, and which expresses the epidermal stem cell factor in the basal layers of its interadnexal epidermis and follicular epithelium. Also disclosed are methods concerning the administration to the transgenic mouse of a compound effective to treat the skin response.
