Described herein are methods and materials for promoting the acceptance of an allograft in a recipient. The invention relates to the use of a blockade of costimulation and hematopoietic stem cells in an allograft transplantation for a recipient mammal receiving a graft from a donor mammal of the same species. Specific properties of the inhibitor of the CD40 ligand-CD40 costimulatory interaction are provided.
