Hematopoietic cell method for treatment of HIV infection

O’Donnell, Jr., Francis E.; St. Louis, MO, USA

Described herein are methods of preparing host cells, namely, autologous lymphocytes, which are devoid of infrastructure and required for viral replication, by treating the cells in vitro, with a multi-drug array of antivirals and/or ionizing radiation in conjunction with chemotherapy. This invention addresses the risk of drug resistance in HIV infection and proposes a reduction of such resistance by suppression of the viral load using novel hematopoietic cells. Methods are provided by which modified CD4 lymphocyte host cells are used to capture virions in a sterile micro-environment, such that the host’s CD4 T-cell lymphocytes are replaced with lymphocytes derived from autologous or homologous stem cells which do not express the CKR-5 receptor, thereby further inhibiting viral load.
