This invention describes methods for producing or obtaining gene targeting constructs via homologous recombination in host cells, as well as the resulting targeting constructs per se. The invention also describes transgenic animals having targeted mutations introduced in to the cells of the animal according to this method. Further disclosed are means of preparing shuttle vectors for the transformation of yeast cells and homologous recombination, and isolation of the targeting vectors from this process.
