This invention introduces a recombinant-based method for increasing the efficiency of genomic targeting procedures in a cloned mammal. Described herein is a two-vector system which introduces vectors into a bacterial cell such that homologous recombination between the vectors occurs. Experimental applications are included for the generation of mutations in a recombination-based construction. Further provided are methods for incorporating such mutations into mouse embryonic stem cells and transgenic mice.
