Described herein are compositions of stem cell populations, methods and uses thereof. The invention relates to an ex vivo process by which pluripotent cells are expanded and optionally transfected to induce quiescence. Claims of the invention include improved long-term engraftment. Therapeutic applications in transplantation and gene therapy are provided.
