Early stage multipotential stem cells in colonies of bone marrow stromal cells

This invention describes a population of cells comprising two classes of bone marrow stem cells (MSCs), one of which are small and rapidly self-renewing stem (RS) cells, and the other of which are large, more mature marrow stromal cells (mMSCs). Described herein are adult bone marrow stromal cells (MSCs) that can differentiate into multiple non-hematopoietic cell lineages. Colonies of human MSCs have been shown to contain both the small, rapidly self-renewing stem cells (RS cells) and the large, more mature cells (mMSCs), enriched samples of which exhibited a greater potential for multipotent differentiation from the RS cells than from the mMSCs. The RS cells were also shown to have a series of surface epitopes and expressed proteins that can be used to differentiate RS cells from mMSCs. Results are described suggesting important distinguishing features between the two major sub-populations of MSCs in defining their biological and clinical potentials for cell and gene therapy.
