Described herein are methods of producing an immune system suppressor and cell stimulator. The invention relates to a therapeutic process incorporating immune system suppressor cells and immune system stimulator cells from a combination of stem cells and lymphoid-derived cells to which a lipopolysaccharide is added as well as a factor selected from the group consisting of a granulocyte macrophage-colony stimulating factor, a macrophage colony stimulating factor, and a granulocyte-colony stimulating factor. Methods of administering such immune system suppressor and stimulator cells from the co-culture into a host are provided. Therapeutic applications are included, such as in the promotion of allograft survival via the co-culturing of stems cells with lymphoid-derived cells.
