This invention describes the use of homologous recombination to inactivate genes associated with antigenic expression, thereby yielding a cell line that may be used as allogeneic donor cells. The genetic engineering ex vivo of the mammalian cell line yields a diminished rejection response due to the lack of at least one functional Major Histocompatibility Complex (MHC) antigen. Methods for reducing and eliminating the expression of these Class 1 MHC antigens, as well as the precise methods by which homologous recombination is applied to the non-transformed mammalian somatic cells, are also described.
