Described herein are novel neural stem cell cultures, methods and uses thereof. The invention relates to the isolation, characterization, proliferation, differentiation and transplantation of mammalian neural stem cells in a culture medium which has been supplemented with specific combinations of supplemental agents. Constituents of the culture medium and their proportions are disclosed, as are methods of their preparation. Therapeutic applications are included for the treatment of epilepsy, stroke, Huntington’s disease, Alzheimer’s disease, multiple sclerosis and various neuropathies.
