Described herein is a hematopoietic stem cell proliferation agent containing IGF-I, and its uses in the culturing of hematopoietic stem cells. The invention relates to a method of culturing hematopoietic stem cells in a medium containing IGF-I and at least one protein selected from the group consisting of SCF and M-CSF. Further disclosed are methods of proliferating the hematopoietic stem cells in a mammal and administering IGF-I with at least one protein selected from the group consisting of M-CSF and G-CSF to the mammal, in an amount sufficient to stimulate hematopoietic stem cell proliferation.
