This invention describes a novel system for producing pancreatic islet cells derived from embryonic stem cells. Methods are disclosed by which differentiation is initiated towards endoderm cells, and focused using reagents that promote the emergence of islet precursors and mature insulin-secreting cells. Also described are methods by which high quality populations of islet cells can be produced in commercial quantities for use in research, drug screening, or regenerative medicine, especially in the treatment of conditions associated with a deficiency of insulin, glucagon or somatostatin such as type I diabetes.
