High dose radionuclide complexes for bone marrow suppression

This invention describes a method for the suppression of bone marrow (BM) and the treatment of conditions that arise in or near bone such as cancer, myeloproliferative diseases, autoimmune diseases, infectious diseases, metabolic diseases or genetic diseases, with compositions having as their active ingredient a radionuclide complexed with a chelating agent such as macrocyclic aminophosphonic acid. Further disclosed are methods for hydrating the patient, as well as methods for administering a dose of 166 Ho-DOTMP to the patient in an aqueous vehicle comprising an effective antiradiolytic amount of a pharmaceutically acceptable radioprotectant, such that the dose delivers approximately 20-60 Gy to the bone marrow of the patient, which is followed by the administration of at least 200 mg/m2 melphalan to the patient, after which the patient is provided with an autologous stem cell transplant, thereby avoiding subjection to total body irradiation.
