This invention describes methods of generating the lambda (lambd) or P1 bacteriophage vectors that are useful in the targeted mutagenesis of eukaryotic cells and in the expression of genes and proteins. Also disclosed are methods for the identification of a lambd or P1 bacteriophage vector having a desired nucleic acid chosen from an assortment or library of bacteriophages, each of which possess a different nucleic acid insert. Further provided are methods for using such vectors in gene targeting and in the expression of genes and protein, as well as the precise conditions and procedures for growth, recombination and identification of the bacteriophage.
